Abstract
The efficient delivery of natural bioactive compounds remains a major challenge due to their poor aqueous solubility, volatility, and limited stability. This study details the fabrication of an antibacterial system based on polycyclodextrin (PCD) network anchored to the surface of silver nanoparticles (NPs) (Ag/PCD nanostructure (NS)) for efficient loading and controlled delivery of carvacrol (CV), as a natural bioactive agent (Ag/PCD@CV). This system was characterized using FT-IR, XRD, TGA/DSC, FE-SEM, EDX mapping, zeta potential analysis and UV-Vis spectroscopy. In vitro release studies exhibited a sustained CV release behavior (~72%) in Ag/PCD@CV NS over 48 h. Free radical scavenging activity increased from 21.6 ± 3.4% in Ag NPs to 73.5 ± 3.4% in Ag/PCD@CV, as measured by the DPPH assay, implying strong antioxidant potential of CV. The MTT assay using L929 fibroblasts demonstrated a reduction in the cytotoxicity of Ag NPs following PCD coating, while Ag/PCD@CV exhibited a cell viability of 72.3 ± 2.4% under the studied conditions. Simultaneously, Ag/PCD@CV demonstrated enhanced antibacterial efficacy, showing approximately 56% greater inhibition against Staphylococcus aureus (S. aureus), compared to Ag/PCD. Overall, the results presented in this work demonstrate the potential of Ag/PCD@CV as a multifunctional bioactive delivery system, exhibiting sustained CV release, antioxidant activity, good cell viability, and enhanced antibacterial activity against S. aureus. These findings support further investigation of Ag/PCD@CV NS as a bioactive platform with antibacterial potential.
